You want to be working ahead of time and drip feeding the tweets out so you can focus on promoting the account, while core content continues to get posted on autopilot. So a few percentages here of content that gets drip fed 40% is just generic content. 20% will be your week 20% will be your retweet bait and then 40% will be interesting links.
For your generic content, this is going to be general content your followers might be interested in. This should define your brand, aka the reason people are following you You want to revise quote source material and tweak well to perform well on Twitter to write good to write a good tweet, think like a tabloid writer. Use headlines to get attention and get people reading. Notice. Notice startling choice of words, use of intrigue and extreme specificity. And then aim for 120 characters and use hashtags liberally.
So use hashtags as much as you want to. For your retweet bait. This is how Twitter users will discover you. It's also how you will increase your influence on Twitter. Being retweeted is the main metric of credibility. There's really an art and science of getting retweeted.

Okay, so just google Link shortening service and you'll see what I'm talking about. And then tweak links to your own site, but to other sites as well. Don't just use the headline, Twitter as a headline engine. So test different headlines as well.
